Mapping QTLs for response to Cadmium stress in Populus nigra

Riassunto: The increasing number of sites contaminated by heavy metals causes several management problems and often leads to a waste of natural resources, such as soil and water, which cannot be utilized. Biological techniques for remediation of polluted soils can give acceptable results with a low environmental impact, even in the long term. Recently, several researches have been conducted on the use of trees from genus Populus for phytoremediation Actually, poplar possesses genetic resources for the identification of genotypes with high tolerance to heavy metals and a good capacity for uptake and accumulation of contaminants. The study reported here focuses on the genetic basis of Cd tolerance in Populus nigra. Two P. nigra genotypes, belonging to natural Italian populations from contrasting geographic and environmental origins, were crossed to create full-sib progenies. Ninety-two F1 individuals were analyzed to construct genetic maps based on the pseudo-testcross strategy. From previous experiments it was also observed that the two parental genotypes presented different tolerance to Cd stress. A large scale experiment with Cd-stress and control was carried out on 92 F1 plants of the mapping progeny for QTL analysis (four replicates per genotype and per treatment, either Cd stress or control). Ecophysiological and chemical parameters were measured during the experiment and used for the genetic dissection of plant response to Cd. Correlations among allelic variants and the phenotypic traits were calculated to identify QTLs controlling responses to Cd stress and Cd accumulation in plant organs (root, stem, leaf). Thirty one QTLs were identified for the female parent 58-861 and 28 for the male parent Poli. Among them, 12 QTLs on 58-861 map and 8 on Poli map co-localized on the linkage group IV. Therefore, this chromosome could contain major genes for the response to Cd stress. Transposition of the common map interval of these QTLs on the Populus trichocarpa genome could allow the identification of these genes. This could be the first step for the development of marker assisted selection of poplar material suitable for phytoremediation.
