Phylogeography of Quercus cerris L. (Fagaceae) based on cpDNA markers

Riassunto: Quercus cerris L. (Turkey oak) is a deciduous oak native to southern Europe and Asia Minor, which belongs to subgenus Cerris. It is very common at medium elevation in peninsular Italy and Greece, and is also found as scattered groups over large areas with sub-continental climate throughout the Balkans. The Tertiary and early Quaternary fossil pollen found in several European countries did not allow taxonomic identification at the species level within the Cerris group. However several remains have been identified in the Balkans. We performed a phylogeographic study based on the analysis of cpDNA on 140 populations collected over the whole range. We amplified six microsatellite regions of the chloroplast genome utilising âuniversal’ primers, that resulted to be polymorphic. Results showed: i) an important level of genetic divergence between the Italian and the eastern populations; ii) a high haplotypic diversity within the Balkan area; iii) the presence of unique Balkan haplotypes. Moreover, Q. cerris shares the two most common Italian haplotypes with Q. suber thus indicating a possible unidirectional hybridisation/introgression between turkey oak (as seed parent) and cork oak (pollen parent). As already described for Q. ilex and Q. suber, introgressive hybridisation should be an ancient event resulting by repeated backcrosses that led to a complete haplotype sharing between the two related species. The haplotype diversity detected in the eastern part of the range, and particularly in the Balkans, suggests this region as a biodiversity hotspot for the species.
